The Roche Innovation Center Zurich is active in the research and development of engineered therapeutic antibodies, with a focus on the fields of cancer immunotherapy and immunotherapy in general. Part of the Roche Group since 2005, it employs about 180 people.

The Position:

The candidate will join the Cancer Immunotherapy (CIT) 2 Department at Roche Innovation Center Zürich (Schlieren, Switzerland) and will interact closely with the team of scientists and research associates working on in depth characterization of tumours and development of novel class of cancer immunotherapy agents.

Primary human tumor explants are considered as one of the most relevant pre-clinical tumor models to assess the activity of portfolio assets. Therefore, the candidate will focus on the optimization of ex vivo primary human tumor explant models and assessing of their predictive potential of clinical response.

This is a collaborative project for 2 years sponsored by the Center of Excellence for Human Translational Bioengineering (HTB). The position is temporary for 1 year with the option to extend to 2 years.

The principal responsibilities will include:

·  Comparison of different systems enabling ex vivo tumor explant cultures in 3D, improvement of conditions to enable longer culture times and increase of viability of tumor, stroma and immune cells
·  Processing of untreated and treated primary human tumor and blood samples for downstream analysis, including the assessment of tumor cell lysis, immune cell activation and proliferation using the following technologies: FACS sorting, multiparametic flow cytometry (> 12 colors) or CyTOF, single cell RNA-sequencing (scRNA-seq), LDH release, IHC and Imaging, CBA
·  Managing of one Research Associate, planning, organization and coordination of team work and experimental activities
·  Data analysis and interpretation, preparation of scientific presentations, regular presentations at internal and external meetings
·  Writing of scientific reports, patents and research articles
·  Build a cross-department network of colleagues working on Human Translational Bioengineering projects

For this position you bring the following qualifications:

·  PhD in tumor immunology, cell biology or cancer immunotherapy
·  Excellent hands-on experience with ex vivo human primary 2D and 3D tumor tissue culture models or organoids and their application for drug assessment
·  Excellent hands-on experience in state-of-the art immunoprofiling techniques for functional characterization of immune responses (FACS-based cell sorting, multiparametric FACS analysis of tumor and immune cells is a must; experience with single RNA sequencing, CyTOF, IF, imaging, spatial-omics and high dimensional data analysis is a plus)
·  Excellent written and verbal English is mandatory
·  Strong inter-personal, communication and presentation skills
·  Ability to work effectively and independently within a diverse group of people
·  Excellent organizational skills, high flexibility and the ability to take initiative and work according to time-lines
·  Maintenance of the laboratory/workstation in accordance with current technical, safety and regulatory standards
